- It may seem obvious that a good night's sleep will be beneficial to motor function in both healthy individuals and those with Parkinson's Disease. However, it is not known to what extent poor sleep affects motor activities in PD. Most patients are aware that a good night's sleep or a nap results in improvement in the symptoms of PD. The sleep benefit phenomenon suggests that dopamine synthesis and storage increases during sleep in the remaining dopaminergic nerve terminals. On the other hand, there is some evidence that sleep benefit in patients with Parkinson's Disease is independent of sleep quality, or even associated with more disturbed sleep compared to findings in patients without sleep benefit.
- Animal research provides evidence that dopamine interferes with cycles of sleep wakefulness and that sleep deprivation influences the dopamine system. In normal rats, selective rapid eye movement (REM) sleep deprivation increases behavioral (sic) and motor responses to dopamine agonists such as apomorphine, an effect possibly mediated by supersensitivity of postsynaptic dopamine receptors. REM sleep is that component of normal sleep in which a person experiences dreams. Researchers can detect these cycles of sleep by recording eye movements along with the brain waves. In Parkinsonian rat models, a significant increase in ambulatory behavior (sic) and rearing in animals with bilateral lesions of the nigrostriatal tract was seen after 3 days of selective REM sleep deprivation.
- REM sleep deprivation also induces a clear increase in apomorphine-induced turning behavior (sic) in rats with unilateral 6-OHDA nigrostriatal lesions, suggesting increased supersensitivity of dopamine receptors.
